The Victorian Centre for Functional Genomics at the Peter MacCallum Cancer Centre is accessible to biomedical researchers Australia-wide and encompasses genome-wide siRNA, miRNA and pooled shRNA screening platforms. The facility houses the human and mouse whole genome siRNA SMARTpool and duplex reagents, the lentiviral based pGIPZ human shRNA-mir library and the human miRNA mimic and inhibitor collection. The facility has been in operation since late 2007 and generates high titer virus for the shRNA whole genome collection, offered as complex pools of 4608 constructs or, as smaller boutique pools for specific library collections (e.g apoptosis, cell cycle, polarity and invasion and kinases). We also house the mouse whole genome TRC collection from Sigma.
As a facility we have screeners with diverse interests, although the majority are primarily focused on cancer. We have a fully equipped high throughput automation platform and perform high content imaging screens using the Cellomics Arrayscan and CellInsight instruments. The facility operates on a researcher driven, staff assisted policy, and provides SOPs and workflows for all aspects of high throughput siRNA/miRNA screening, viral production and transduction. We have a dedicated bioinformatician that works alongside the screeners on all aspects of screen analysis.
